Liquid Sporulation

1. Grow cells overnight in YEPD
2. Remove 200ml; add 5ml water
3. Spin 5'
4. r/s pellet in 2.5ml SPM
5. incubate at 25°C with good aeration for 2-4 days
6. Take 1ml, r/s in 100ml zymolase -- dissect
